6-ROX是一种用于在实时定量PCR中的荧光报告基因信号进行标准化的ROX参比染料,与其他罗丹明染料相比,6-ROX非常不稳定。但经改良后的6-ROX glycine具有较好的稳定性。6-ROX glycine具有与6-ROX相似的光谱性质。该参比染料是溶于20 mM Tris(pH 8.4),0.1 mM EDTA和0.01%Tween 20中的25 μM溶液。

◆实验结果使用什么仪器观察? 实验结果可以通过流式细胞仪和荧光显微镜进行观察,但更推荐使用流式细胞仪。因为正常的细胞和凋亡的细胞膜上都有磷脂酰丝氨酸(PS),只是凋亡细胞膜外更多,可以结合更多的Annexin V,流式细胞仪更灵敏,可以区分出微小差异,将正常细胞与凋亡细胞分成不同的两群,但如果用荧光显微镜,可能观察不到这一差别,尤其是贴壁细胞。若要在成像测定中检测细胞凋亡,推荐使用SuperView™ 488 Caspase-3 Assay Kit for Live Cells。
