MA qPCR活菌检测试剂盒为检测细菌活性提供有效的手段,本试剂盒提供PMA染料和基于SYBR Green染料的qPCR混合液。染料的最佳用量和可处理的样品数量可能因样品类型而异。PMA是一种高亲和性的DNA结合染料,尤其与双链DNA,该染料本身具有微弱的荧光,但与核酸结合后可以发出更为明亮的荧光。PMA具有细胞膜不可渗透性,因此可选择性地修饰膜受损的死细胞的DNA。PMA修饰的DNA经蓝光(~464 nm)光解后,PMA上的光反应性叠氮基转化为高反应性氮烯自由基,与DNA结合位点附近的任何烃部分反应形成稳定的共价氮碳键,从而导致永久性DNA 修饰。该修饰过程会使DNA不溶,且在后期的基因组DNA提取过程中与细胞碎片一起丢失。残留在溶液中未结合的PMA,在强光照射下与水分子反应分解成无交联活性的羟胺化合物,使其不能再共价结合DNA。基于PMA的这一特性,我司将PMA与qPCR技术相结合,形成一种新的检测方法—PMA-qPCR,用于活细菌的筛选。目前该法已在多种细菌菌株以及酵母、真菌、病毒和寄生虫中得到验证。

产品介绍 Dihydroethidium(Hydroethidine)是常用的 DNA 染料溴化乙锭的还原形式,它可以检测活细胞内的氧化活性,也可以检测吞噬细胞的“呼吸爆发”。Dihydroethidium 本身在细胞中具有蓝色荧光(Ex/Em:355/420 nm),当它进入细胞中,脱氢后会成为氧化形式的乙锭,当与 DNA 嵌合时,氧化形式的乙锭会发出红色荧光(Ex/Em:518/605 nm)。

Calcein AM 是一种对活细胞进行荧光标记的细胞染色试剂,它可以穿透细胞膜进入细胞,被细胞内的酯酶剪切为Calcein。Calcein 带高度负电荷,被滞留在正常细胞的胞质内,发出明亮的绿色荧光。由于其低毒性,Calcein AM 可以用于研究细胞膜的完整性和长期细胞示踪实验。此外,它也可被用于定量活细胞数。Calcein AM 细胞活力检测试剂盒是在细胞活力检测中,用来定量活细胞数的终点检测法。测定所产生的荧光信号与样品中的活细胞数成正比(图1)。
